Alcudacigib CAS# 2660218-70-0

Alcudacigib (ASP1570) is a small-molecule inhibitor of diacylglycerol kinase (DGK) developed to modulate tumor immunosuppression. In preclinical studies, it inhibited DGK activity in vitro and restored T-cell effector functions that were suppressed by immunosuppressive signals, leading to enhanced CD8⁺ T-cell activation. In syngeneic mouse tumor models, oral or systemic alcudacigib treatment induced significant tumor growth inhibition that depended on cytotoxic T-cell engagement, demonstrating antitumor efficacy in immunocompetent hosts. These data support its mechanism as an immune-modulating anticancer agent targeting DGK to overcome T-cell suppression in the tumor microenvironment.
