摘要:本发明公开了一种苯甲酸阿格列汀的制备工艺,通过3‑甲基‑6‑氯尿嘧啶与2‑氰基溴苄在碱性条件下发生亲核取代得到2‑(6‑氯‑3‑甲基‑2,4‑二氧代‑3,4‑二氢‑2H‑嘧啶‑1‑基甲基)‑苄腈(化合1),再与(R)‑3‑氨基哌啶二盐酸取代得(R)‑2‑[(6‑(3‑氨基哌啶‑1‑基)‑3‑甲基‑2,4‑二氧代‑3,4‑二氢嘧啶‑1(2H)‑基)甲基]苄腈(化合物2),最后与苯甲酸成盐取得到苯甲酸阿格列汀(化合物3),每一步目标化合物结构经ms,1H‑NMR确证,通过HPLC测定纯度.该方法原料易得,反应条件温和,操作简便,可以大量生产来满足使用需求,是一种高效绿色环保工艺方法,适用于工业化生产.
