Vamorolone is a dissociative steroid designed to retain anti-inflammatory activity while minimizing the side effects associated with classical glucocorticoids. In vitro, it binds the glucocorticoid receptor with high affinity and suppresses pro-inflammatory gene expression while exhibiting reduced transactivation of genes associated with metabolic and bone-related adverse effects. In vivo, oral administration in rodent models of inflammatory disease at doses of 1–30 mg/kg reduces markers of inflammation, improves muscle strength in dystrophic models, and demonstrates a significantly lower impact on glucose metabolism and bone density compared to prednisone.
