Ozanimod (RPC‑1063) is a highly potent and selective S1P₁ and S1P₅ receptor agonist, with EC₅₀ values of ~1 nM for S1P₁ and ~8–11 nM for S1P₅ in human and animal receptor assays. It induces sustained S1P₁ receptor internalization in vitro and, when administered orally at 0.05–1 mg/kg daily for 14 days in rodents, potently reduces circulating lymphocyte counts and ameliorates disease in preclinical models of autoimmune encephalomyelitis and colitis.
