Sepiapterin is a natural precursor of tetrahydrobiopterin (BH4), a critical cofactor for nitric oxide synthases and aromatic amino acid hydroxylases. In vitro, sepiapterin restores intracellular BH4 and nitric oxide (NO) production in endothelial cells with EC₅₀ values around 1–10 µM, improving eNOS coupling and reducing superoxide formation. It also increases dopamine synthesis in neuronal cultures by enhancing tyrosine hydroxylase activity. In vivo, sepiapterin administration (5–20 mg/kg, i.p. or oral) elevates tissue BH4 levels, improves endothelial function, and reduces blood pressure in hypertensive or diabetic rodent models. In Parkinson’s disease models, sepiapterin boosts striatal dopamine content and mitigates motor deficits. Overall, sepiapterin demonstrates potent biochemical restoration of BH4-dependent enzymatic functions and vascular/neuroprotective efficacy in preclinical systems.
