Envonalkib (TQ-B3139) is a potent, orally bioavailable ALK tyrosine kinase inhibitor with demonstrated efficacy in both in vitro and in vivo models. In preclinical studies, it exhibited high potency against ALK-positive cell lines, with an IC₅₀ of approximately 1.96 nM. In vivo, it significantly inhibited tumor growth and demonstrated improved central nervous system (CNS) penetration compared to crizotinib. Clinically, a Phase III trial involving 264 patients with advanced ALK-positive non-small cell lung cancer (NSCLC) showed that envonalkib achieved a median progression-free survival of 24.87 months, nearly double that of crizotinib (11.60 months), with a confirmed objective response rate of 81.68% versus 70.68% for crizotinib. These findings underscore envonalkib's superior anti-tumor activity and CNS efficacy in treating ALK-positive NSCLC.
