Umbralisib is a dual inhibitor of PI3Kδ (phosphoinositide 3-kinase delta) and casein kinase 1 epsilon (CK1ε), enzymes involved in the regulation of cell survival and proliferation in hematologic malignancies. In vitro, Umbralisib exhibits nanomolar potency against PI3Kδ and effectively suppresses downstream AKT phosphorylation and survival signaling in B-cell lymphoma and leukemia cell lines. Its selectivity for the δ isoform allows for targeted immune modulation with reduced off-target effects compared to pan-PI3K inhibitors.
