PSMA-11 (HBED-CC) is a small-molecule ligand designed for targeting prostate-specific membrane antigen (PSMA) and serves as a chelator for radiolabeling with ^68Ga. In preclinical binding studies, PSMA-11 showed high affinity for PSMA (IC50 ~2 nM) and selective uptake in PSMA-expressing prostate cancer cells, with minimal binding to PSMA-negative cells. In xenograft mouse models, ^68Ga-PSMA-11 demonstrated rapid, high tumor uptake (>10% ID/g within 1 h) and favorable tumor-to-background ratios, with fast renal clearance. Its stability and specificity enabled sensitive detection of small lesions in vivo. These findings validated PSMA-11 as a promising PET imaging agent for prostate cancer diagnostics.
