合成路线:T-1815 is prepared by the condensation of 2-dimethylamino-2-(2-thienyl)-butanol (I) with 3,4-dimethoxybenzylchloride by means of KOH.Starting compound (I) is obtained as follows: the condensation of the isocyano compound (III) with ethyl chloroformate in the presence of lithium diisopropylamide affords ethyl 2-isocyano-2-(2-thienyl)-butyrate (II),which is then reduced with lithium aluminum hydride,followed by treatment with HCOOH/HCHO to give the amino-alcohol (I).
