Viloxazine is a selective norepinephrine reuptake inhibitor (NRI) with additional modulatory effects on serotonergic signaling. In preclinical transporter assays, it inhibited the norepinephrine transporter with an IC50 of ~0.2–0.5 µM and showed minimal affinity for dopamine or serotonin transporters. Viloxazine increased extracellular norepinephrine and, to a lesser extent, serotonin in rodent brain microdialysis studies. It also acted as a 5-HT₂B and 5-HT₂C receptor modulator, contributing to its antidepressant and pro-cognitive profile. In animal behavioral models, viloxazine demonstrated antidepressant- and ADHD-relevant activity, including improved attentional performance. Pharmacokinetic studies showed rapid absorption and good brain penetration, supporting clinical development.
