Remdesivir CAS# 1809249-37-3 瑞德西韦
Remdesivir (GS-5734) is a monophosphoramidate prodrug of an adenosine analog that inhibits viral RNA-dependent RNA polymerase. In preclinical assays, its active metabolite (GS-441524 triphosphate) showed potent inhibition of viral replication, with EC50 values of 0.01–0.1 µM against coronaviruses, including SARS-CoV and MERS-CoV. In human airway epithelial and animal cell cultures, remdesivir caused dose-dependent suppression of viral RNA synthesis. In mouse and rhesus macaque models of SARS-CoV and MERS-CoV infection, prophylactic or early therapeutic administration reduced viral loads and improved lung pathology and survival. Pharmacokinetic studies demonstrated favorable distribution and intracellular activation, supporting its development for RNA virus infections.
