Deutivacaftor CAS# 1413431-07-8

Deutivacaftor (VX-561, CTP-656) is a deuterated analog of ivacaftor developed as a CFTR potentiator with improved metabolic stability and longer half-life. In vitro, deutivacaftor enhances CFTR chloride channel gating in human bronchial epithelial cells carrying G551D/F508del mutations with an EC₅₀ of approximately 255 nM, showing potency comparable to ivacaftor but with slower oxidative metabolism due to deuterium substitution. Pharmacokinetic studies in animals and humans demonstrate high plasma protein binding (>99%) and an extended elimination half-life (~19 h), supporting less frequent dosing.
